In pharmaceutical chemistry, a slight difference in the three-dimensional arrangement of a molecule can lead to significant differences in its biological effects. Chiral drugs are compounds that exist in two non-superimposable mirror-image forms known as enantiomers. Although these enantiomers share the same chemical formula, their biological responses in the human body may differ considerably. This phenomenon arises because biological systems—such as enzymes and receptors—are inherently chiral. As a result, they interact selectively with one enantiomer over the other. In many cases, one enantiomer is primarily responsible for the desired therapeutic effect, while the other may be less active or even associated with adverse side effects. Thalidomide represents a well-known historical example. It was prescribed as a sedative in the mid-20th century; however, one of its enantiomers was linked to severe congenital malformations, leading to stricter global drug regulations. A more recent example is esomeprazole, the optically pure active enantiomer of omeprazole, developed to enhance therapeutic response and reduce variability among patients. Similarly, ibuprofen exhibits differences in activity between its enantiomers, with one form being more pharmacologically active as an anti-inflammatory agent. Misuse of chiral medications—whether through improper dosing, self-medication, or lack of medical supervision—may reduce therapeutic efficacy or increase the risk of side effects. Therefore, modern pharmaceutical industries focus on developing enantiomerically pure drugs rather than racemic mixtures whenever possible. Advanced separation and analytical techniques play a crucial role in ensuring the purity and quality control of chiral drugs, thereby enhancing patient safety and minimizing potential risks. Understanding the nature of chiral pharmaceuticals is essential for promoting rational and safe drug use, and it highlights the importance of scientific research in safeguarding public health. ⸻ College of Science – Al-Mustaqbal University
